Elena ha perso i genitori il 23maggio2009 a causa di un incidente stradale, di cui è l'unica superstite. Stefan le rivela, inoltre, di essere colui che l'ha salvata dall'incidente stradale nel quale sono morti i suoi genitori, e che inoltre non è una Gilbert, perché è stata adottata. Elena scopre che la sua madre naturale è Isobel Fleming, la moglie del professore di storia Alaric Saltzman, che tutti credono morta, ma che in realtà è stata trasformata in vampiro da Damon. Negli ultimi episodi scopre invece che suo padre è colui che aveva sempre creduto essere suo zio, John Gilbert. Da parte di madre, Elena è parente di Katherine e questo motiva la somiglianza tra le due.

Elena deve affrontare nuovi problemi: la sua amica Caroline si è trasformata in un vampiro grazie al sangue di Damon dopo essere stata uccisa da Katherine, mentre Tyler è diventato un lic. Damon le confessa, inoltre, i suoi sentimenti, ma le fa dimenticare tutto con un bacio sulla fronte. L'arrivo di Elijah la rende pronta a morire per coloro che ama. Deve anche affrontare la possibile morte di Damon a causa del morso di Tyler. Katherine Pierce/Katerina Petrova
È la donna che vampirizzò i fratelli Salvatore nel 1864. Sebbene Damon sia convinto che fosse stata rinchiusa in una cripta, Katherine in realtà riuscì a fuggire e a mettersi in salvo per conto proprio, sparendo senza lasciare traccia. Appare in alcuni flashback e torna a Mystic Falls nell'ultimo episodio della prima stagione, per poi diventare a pieno titolo una presenza fissa nella seconda. Il suo vero nome è Katerina Petrova e proviene dalla Bulgaria. Sempre nella seconda stagione si scoprirà tutta la sua storia e il vero motivo per cui è tornata a Mystic Falls.
